how to simulate for the LO-RF isolation for a mixer using spectreRF??

You can simulate using pss+pxf. check out the cadence documentation in you intstallation path for details.
You can also do a simpler/quicker sp analysis for get a idea of the LO to RF feedthru.

Please bear in mind though, an ideal schematic simulation often will give an unrealistic performance of LO to RF leakage because effects which dominate this (for espically differential circuits) are layout and mismatch. So post extraction simulation in addition to monte-carlo is needed to get a realistic figure.
