There is something wrong in your settings as far as I can read:

No, this is wrong, the AMSHOME must be pointing to the IUS install directory rather. Besides, you do not need to miss about any of the hdl.var, cds.lib files. They should be working fine as provided.
To get yourself sorted out you need to:
1. Start a new Unix terminal
2. Delete your run directory and start from a new unziped/untared archive. Leave all the files as provided
3. Acquire the IC stream, i.e. where the cadence DFII is installed
4. Acquire the IUS stream, i.e. where ncvlog, ncelab .. are installed.
5. Updated your LD_LIBRARY_PATH to get the $AMSHOME/tools/lib added.
6. run the run-me-with-gui script.
All should be fine if you go carefully through the above steps.
Please give it a try and let me know.
BTW, are you a RHEL or a SUSE user ?
